What does Niacin do for the brain?
Niacin (Vitamin B3) is the precursor to NAD+, the coenzyme required for mitochondrial ATP production. Neurons consume roughly 20% of the body’s resting energy on 2% of body mass, making them highly dependent on adequate NAD+. Since NAD+ levels decline measurably with age, niacin supplementation supports the substrate side of cellular energy production. Niacin also causes microvascular dilation, improving capillary-level nutrient delivery.

Why energy is the bottleneck
There is a sequencing problem in cognitive supplementation that most formulas ignore. Increasing blood flow to a neuron delivers more oxygen and glucose. But converting oxygen and glucose into usable energy requires functioning mitochondrial machinery, and that machinery requires NAD+.
Deliver fuel to an engine that cannot burn it and you have achieved nothing. This is the specific reason niacin is in the formula, and it is arguably the ingredient that makes the other five worth including.
What NAD+ actually does
Nicotinamide adenine dinucleotide is a coenzyme present in every cell. Its function is electron transfer — it accepts electrons during the breakdown of glucose and fatty acids, then delivers them into the mitochondrial electron transport chain where the energy is captured as ATP.
Without adequate NAD+, oxidative phosphorylation slows. Less ATP is produced from the same substrate. In a cell with modest energy demands this is tolerable. In a neuron it is not.
The brain is approximately two percent of body mass and consumes roughly twenty percent of resting metabolic energy. Neurons maintain electrochemical gradients continuously, fire repeatedly, and sustain long-term potentiation — the cellular basis of memory formation. All of this is ATP-expensive.
The age-related decline
Declining NAD+ with age is one of the more consistently replicated findings in ageing biology. Multiple mechanisms contribute: reduced synthesis, increased consumption by DNA repair enzymes responding to accumulated damage, and increased activity of NAD+-consuming enzymes generally.
The functional consequence in neural tissue is an energy constraint precisely where it is least tolerable. Slower firing, weakened signal maintenance, and impaired long-term potentiation all follow from insufficient ATP.
This is why NAD+ has become a substantial focus in ageing research, and why supplying its precursor is a coherent supplementation strategy rather than a speculative one.
Niacin as precursor
The body synthesises NAD+ from several precursors, of which niacin is the most established and the most extensively documented. The NIH Office of Dietary Supplements maintains detailed fact sheets on niacin intake requirements and metabolism.
Niacin has one of the longest safety records of any vitamin, having been used at pharmacological doses in lipid management for decades. Its metabolism, interactions and side effect profile are unusually well characterised.
The flush, explained properly
Niacin causes prostaglandin D2 release from skin cells, which dilates cutaneous capillaries. The result is warmth, sometimes visible reddening, occasionally mild itching, across the face, neck and upper chest.
It begins fifteen to thirty minutes after dosing, lasts twenty to forty minutes, and diminishes substantially with continued use as tolerance to the prostaglandin response develops.
Three things worth knowing. It is completely harmless. It is not an allergic reaction. And it is not an indicator that the product is working — many people never experience it at all and derive the same benefit. Taking the capsule with a small amount of food reduces it considerably.
The secondary vascular benefit
The same vasodilation that produces the flush has a genuine functional value. Niacin dilates microvasculature — the smallest capillaries, where actual exchange between blood and tissue occurs.
The arginine and citrulline group dilates larger vessels, increasing bulk delivery. Niacin dilates the terminal capillaries, ensuring that delivery actually reaches tissue. The two operate at different points in the vascular tree and are complementary rather than redundant.
Frequently asked questions
Is the niacin flush harmful?
No. It is a prostaglandin-mediated cutaneous vasodilation with no tissue consequence. It typically diminishes within the first one to two weeks.
Can I avoid the flush?
Taking the capsule with a small amount of non-protein food reduces it substantially, as does avoiding alcohol and hot beverages in the hour after dosing.
Does niacin interact with statins?
Niacin at higher doses has been associated with increased myopathy risk in combination with statins. If you take a statin, discuss this with your physician before starting.
Is niacinamide the same thing?
No. Niacinamide (nicotinamide) is a different form that does not cause flushing but also lacks niacin's lipid and vasodilatory effects. They are not interchangeable.
